肉毒桿菌神經毒素（BoNT）是人類已知的毒性大的物質之一。已經鑒定了七種免疫學上不同的神經毒素血清型，命名為A至G型（BoNT / A，BoNT / B，BoNT / C，BoNT / D，BoNT / E，BoNT / F，BoNT / G）。這些肉毒桿菌神經毒素天然產生與一種或多種無毒神經毒素相關蛋白（NAP）復合。NAP對于攝入毒性很重要，因為它們保護神經毒素免受蛋白酶，酸性，熱的影響，并且它們也可能在粘膜層的易位中發揮作用。
合成為單個150kDa多肽鏈，隨后通過切割激活復合物的實際神經毒素部分以產生兩條鏈，重鏈和輕鏈，其通過單個二硫鍵連接。對于每種毒素，50kDa輕鏈是鋅??依賴性蛋白酶，其切割神經傳遞期間突觸小泡膜融合所必需的單個靶蛋白。A，C和E型神經毒素特異性結合并選擇性地切割突觸體相關蛋白SNAP-25，而B，D，F和G型切割突觸小泡蛋白-2，也稱為VAMP-2。靶蛋白的切割抑制神經元之間的神經遞質釋放，這導致肌肉麻痹。
